大唐垂拱年间，狄仁杰（冯绍峰 饰）因不满武后（邬君梅 饰）为野心牺牲无辜，一行人离开神都，巡查江南道。不想昭昭盛世之下，却是妖鬼横行。权力的倾轧、利欲的贪糜、执念的癫狂将狄仁杰拉入了一个个精心编织的弥天大网之中。
剧集采用单元案件串联主线的模式，围绕狐爪伤痕、水鬼索命、傩戏泣血、巨灵流泪、古寺婴啼五大奇案展开。狄仁杰在巡查江南途中，与段十四（李彧 饰）、楚青岚（邵芸 饰）、林不言（单敬尧 饰）组成探案团队，以近妖之智探微毫，以天人之技窥人心，在迷雾交织的诡事中抽丝剥茧，侦破连环奇案，逐渐触及朝堂深处一只翻云覆雨之手。
